Heroes are always playable, you can't lose them. The choice is to take him to prison in a town that has one or to leave him on the map under a tombstone available for his army to come back and dig up to revive. I suppose you could leave him on the map and leave a force to guard him but we won't know if that is worthwhile until the game is released.

I would think if I am going to leave an army to guard a hero, I would want to do it in one of my towns. First, the defensive bonuses from Castle and towers. Second, If you leave an army on the map, you use up one of your 8 available armies. If you leave the forces in your garrison, it won't do this. Just my thoughts.

questions about the prison
1.Do u need a troop to send the lost enemy hero to your prison or they will sent to the nearest prison automatically?
2.Can any troop,either friend or enemy,get a headstone leaved on the land?
3.Can u free your prisoner if u want?

Imprisonment/Tombstoning occurs when the Hero you killed would have returned to the Pool of Heroes Available for Recruitment in Heroes 3. So instead of returning to the Pool the Hero remains loyal to the Player who hired him/her.
You still have the options of Fleeing and Surrendering, and if these are chosen then there's no Tombstoning / Imprisonment.
As for the questions, we don't really know but a guess would be:
1. No, it probably occurs automatically and instantly.
2. Only Heroes gets headstones. (It's essentially meaningless to flee or surrender if your army contain no Heroes, and likely it will not be allowed. You can't resurrect killed monsters, only heroes.)
3. We don't know.
You can check out the Captives thread I created a while ago which discussed Imrisonment. I was wishing for some advanced options for Imprisonment in it, Niteshade was of another opinion than me as I remember it.
